Abstract

PL35 glycosaminoglycan lyases act on structurally distinct substrates, creating an opportunity to examine how mutations affect enzyme activity across polymers. This study developed a framework for comparing catalytic-site and substrate-binding-site variants of GAGases II, III, and VII using publicly available activity-assay and chromatographic records. Each variant is compared with its corresponding wild type for hyaluronan, chondroitin sulfate, heparan sulfate, or alginate where measurements are available. Relative activity patterns and product records are considered together to distinguish broadly reduced activity from substrate-dependent responses. The framework also separates comparisons supported by available measurements from kinetic parameters that would require substrate-concentration experiments. Because the numerical tables prepared earlier include synthetic values, they illustrate the proposed comparisons but do not establish experimental mutation effects. Reanalysis of the original records, including assay controls, normalization, and replicate variation, is necessary before reporting empirical findings. This approach can guide the identification of variants for further structural and biochemical investigation.
